Abstract

The objective of our project is to contribute to the field of cryopreservation of epididymal sperm and to define the particularities of species in order to facilitate the successful completion of cryopreservation and the establishment of a cryogenic bank of local animal populations threatened with extinction, by the growing infusion of more efficient exotic breeds or those threatened by environmental factors. Three aspects were addressed before contributing to the creation of a national epididymal sperm bank:
The first experiment was to examine for the first time the characteristics and preservation of post-mortem spermatozoa, obtained from the epididymis and vas deferens of adult roosters from the local population. Compare the quality of sperm at the level of the epididymis and the vas deferens. To determine the effectiveness of two post-mortem sperm collection techniques and the effect of in situ storage at different temperatures (2h and 24h at 20 ° C and after refrigeration at 4 ° C for 24h).
The second experiment made it possible to evaluate the quality in pre-freezing and post-freezing of the epididymal sperm of the local goat, as a function of the storage temperature of the testes, and the time elapsed between the death of the animal and the recovery of the sperm (0, 24, 48.72 h), to establish the optimal protocols for cryopreservation of epididymal sperm in this species.
In a third part, a comparison of the effect of 5 µg / ml of three antioxidants in the local bull epididymal sperm freezing diluent: Quercetin, trolox and tannic acid.
According to the results obtained, it can be concluded that good quality semen samples can be collected from the vas deferens with the flushing method, and that the local rooster sperm can be stored in situ at 20°C for 24 hours without change of sperm quality. Room temperature (21.5°C) and refrigeration (4°C) allow maintaining up to 48 hours after death, a very good quality of goat epididymal semen before freezing. Cryopreservation of the goat's epididymal sperm stored in situ for 48 hours at 4 ° C did not significantly influence the sperm parameters: motility, progressive mobility, viability, acrosomal integrity, integrity of the sperm. Membrane and the rate of abnormalities. The addition of the antioxidants to the diluents demonstrated that there is a large significant difference (p <0.001) between the control and the other antioxidants while there is no difference between the three antioxidants. With better values in the two-dilution media with trolox and quercetin for all values in the bovine species.
Finally, 1879 doses of sperm were stored with 41 donors of local breeds (cattle, goats, sheep, and roosters) as a first step in the establishment of an epididymal sperm genetic bank. The difficulties encountered and the recommendations for the success of our project to set up a cryobank for local animal populations are presented.
